Which cycle semi-log paper should I use, and how do I label the Y-axis?

I have just done SDS-PAGE and need to use the gel to plot the distance migrated against log of the molecular weight. The molecular weights of the standard range from 6,000Da – 200,000Da. Which cycle semi-log paper should I be using to plot this? I really am unsure of how I should be labelling the Y-axis. Thank you x

If you ignore the thousands for the moment, it’s a plot from 6 to 200. With the logs being base ten, six falls in the range of 0 to 1 (log 1 = 0, log10 = 1) and two hundred falls in the range 2 to 3 (log 100 = 2, log 1000 = 3) So you’ll need three cycles to cover from 6 to 200 and the same is true for 6k to 200k. (Imagine k is just another bit of the unit – kDa).

The paper should have lines for every whole number in each cycle, so you won’t need to interpolate between them if you have numbers rounded to 1 sig. fig. Don’t get thrown by the fact that they are not evenly spaced – they’re just 1, 2, 3, 4…etc. There can be no y = 0, the x-axis of conventional graphs as there is potentially an infinity of cycles in both directions – larger and larger, and smaller and smaller.

With two sig. figs you will need to interpolate, so imagine the space between successive lines representing 1 sig. fig. are chopped up in the same pattern as each individual cycle of the semi-log paper is chopped up. It is very much a guess, unless you want to get out the log tables and find a figure to be used as if the scale of the y-axis were linear. This way of working actually allows you to do a semi-log (or even a log-log ) plot using ordinary graph paper. Cool, eh?
